Design: The genomic DNA fragments of N.gregoryi SP2 were extracted and then separated by agarose gel electrophoresis with 1% gel. The TruSeq DNA Library Preparation Kits were used to prepare DNA libraries with insert sizes from 400-500 bp for paired-end sequencing. Blunt-end DNA fragments were generated using a combination of fill-in reactions and exonuclease activity. An 'A'-base was then added to the blunt ends of each strand, preparing them for ligation to the sequencing adapters. Each adapter contains a 'T'-base overhang on the 3'-end, providing a complementary overhang for ligating the adapter to the A-tailed fragmented DNA. These adapters contain the full complement of sequencing primer hybridization sites for paired-end reads. Following the denaturation and amplification steps, libraries can be pooled for sequencing.

The Natronobacterium gregoryi Argonaute (NgAgo)-guide DNA (gDNA) system was reported to have powerful genome editing capacity by Han Chunyu group. However, many related studies have shown no genome editing. For example, recently Liang X. group firstly demonstrated the potential of NgAgo/gDNA in inhibiting HBV replication through accelerating pgRNA degradation, but not DNA editing. Therefore, we grew this archaea and then sequenced the whole genome. The existing submissions are not updated for one year already. We tried to analyse the endogenous oligonucleotides-guide DNA and locate it in the genome. Here we sequenced the whole genome of Natronobacterium gregoryi SP2.
